Disulfiram like reaction medications or anti-seizure drugs, but the FDA and others have warned that their overuse can cause serious liver problems. The researchers believe that by targeting and selectively killing Bd Bz genes, they have found the key to treating Huntington's. "Our approach is novel in that it directly inhibits Bz in the liver of mice with Huntington's disease," says lead author Dr. Jeffrey D. Young. "Our results show that the drug can dramatically reduce liver burden of Bd and Bz, which is a very important step towards treating these deadly forms of the disease," writes Dr. Young in an article about the work published today in Journal of Neural Transmission. The study is first to directly target both a Bz gene and Bd in Huntington's disease, the first to show that Bz inhibitors can be safe and effective in human clinical trials. Scientists and doctors have studied ways to selectively kill the Bz genes or their messenger RNA for more than 20 years. In 2001 a team at the University of Washington developed a drug that specifically cleaves the Bz gene, but it was never approved by the government and is no longer available. Drug development has been hampered for almost 20 years because Bz proteins are too complex to target with a single drug. This is one reason why the United States does not currently have a drug to treat Huntington's. In addition, the mice treated with Bbq anti-Bz antibodies showed lower concentrations of Bbq protein in their livers than did the control mice. That indicates targeting both genes may be useful to treat a range of conditions. And the drugs could be even safer and more effective than the mnemonic for drugs causing disulfiram like reaction drugs used to fight malaria because the antimalarial drugs need to be injected, while both the Bbq and Bz proteins can be excreted through the kidneys. "This is a landmark discovery," says Dr. Daniel T. Schwartz, chairman of neurology at the University Pennsylvania's Perelman School of Medicine. "It's extremely exciting to see a single drug work in four different models of Huntington's."
